Congratulations to Soizic Garaud & the LBAI - B Lymphocytes, Autoimmunity and Immunotherapies - Inserm U1227 team at Brest University, for the incredible work developing a spatial analysis pipeline using Phenoplex on IMC data from their Hyperion system (Standard BioTools).
The purpose of the study was to investigate immune infiltration of patient skin samples affected by cutaneous immune-related adverse events (irAEs) due to Immune Checkpoint Inhibitors (ICI), ICI-induced lichen planus, and compare these to control non-drug related lichen planus patient skin samples.
The team employed Phenoplex for tissue and cell segmentation, phenotyping, involving setting thresholds for marker positivity and visual data exploration and QC using the built-in interactive visualization tools. Analysis with Phenoplex results uncovered that ICI-induced lichen planus showed a reduction in T cell infiltration and an increase in B cell frequency, and that these B cells are organized in aggregates close to T cells in the inflamed tissue microenvironment.
